Potential microbial hazards in actual cosmetology trends - bio, vegan, raw

Bio-, vegan- and raw-cosmetics are continually more popular type of cosmetic products caused by an absence of synthetic and preservative substances but at the same time there is an increased probability of microbial contamination. The main aim of this thesis is a practical findings of potential microbiological contamination using a challenge test and a determination of any active ingredients in the cosmetic preparations. The legislation of cosmetic preparations as well as the information about certificates, which are awarded for bio-, vegan- and raw-cosmetics has been processed in the theory. This section also discusses a composition of vegetable and essential oils, which are used as raw materials in cosmetics production to a great extent and have a positive effect on skin. This thesis also deals with a possible microbiological contamination, listing the most frequently occurred pathogenic microorganisms, which can have a negative influence on consumer health. Operating life of selected samples using the Schülke Koko test by inoculating selected representatives of bacteria, yeasts and molds (Candida glabrata, Escherichia coli, Pseudomonas aeruginosa and Aspergillus niger) are being tested in the practical part. In addition a possible antimicrobial activity which may partially replace an effect of synthetic preservatives in the presence of essential oils is also being tested. Finally any biological active substances were analysed by the gas chromatography, in particular fatty acids contained in the creams.
Metodika dezinfekce fotografických a filmových materiálů s želatinovou světlocitlivou vrstvou

Uvedená metodika navrhuje postup identifikace historické fotografické techniky a kinofilmů na acetátové a polyesterové podložce, optimální odběrový materiál a techniku odběru vzorku pro mikrobiologickou analýzu, která vychází z doporučení již publikované „Metodiky odběru a izolace bakterií, kvasinek a plísní z fotografických materiálů“ pomocí suchého stěru pomocí měkké polyuretanové (PUR) houbičky. V případě pozitivního výsledku mikrobiologické analýzy jsou navrženy dezinfekční metody, které nepoškozují černobílé želatinové fotografie (páry n-butanolu) a filmové materiály s želatinovou světlocitlivou vrstvou na acetátové podložce (kvarterní amoniová sůl nebo etylenoxid v plynné směsi) a polyesterové podložce (etylenoxid v plynné směsi).

Metodika dezinfekce fotografických materiálů s kolódiovou světlocitlivou vrstvou

Uvedená metodika navrhuje postup identifikace a následné dezinfekce matných a lesklých kolódiových pozitivů na papírové podložce s barytovou mezivrstvou. Ve svém doporučení mikrobiologického odběru se opírá o již publikovanou „Metodiku odběru a izolace bakterií, kvasinek a plísní z fotografických materiálů“. V případě pozitivního mikrobiologického nálezu jsou v předkládaném textu doporučeny metody (ethylenoxid v plynné směsi, páry n-butanolu), které se ukázaly jako nedestruktivní vůči zkoumaným kolódiovým pozitivům.

V případě pozitivního výsledku mikrobiologické analýzy jsou navrženy dva postupy samotné dezinfekce - expozice v parách n-butylalkoholu nebo ponor do směsi alkoholů Bacillol AF.

Microbial contamination of fruit teas

Teas are consumed daily by millions of people on the World, therefor it is important to care for their microbial harmlessness. The aim of this work was to conduct a reaserch in regards to general information about fruit teas, their manufacturing and possibilities of contamination by undesirable microorganisms. In this research were also described important bacteria and fungii and we focused on mycotoxins and the elimination of microorganisms from a food. Representation of microorganisms in teabags was evaluated in practical part of this research. We focused on total quantity of microorganisms, coliform bacteria, that can be pathogenic in higher representation and micromycetes. Genus structure of micromycetes was important in particular, because they can produce metabolites, mycotoxins, that can be a cause for many diseases. Analyses were conducted by two methods. Variant classified as A was consisting of transformation of the teabag contents into the physiological solution. Variant classified as B was consisting of watering of the teabag contents. During evaluation, both of these variants were compared. From the results of this work it was found out, that quantities of microorganisms were relativem small. It was discovered, that during the sample preparation by watering the sample with boiling water (variant B) the quantity of microorganisms will lower, in some cases they were not present at all. The presence of wider genera of micromycetes was also discovered. Three genera were dominant Aspergillus, Penicillum and Cladosporium. These free found of genera can be dangerous, because of mycotoxins, however micromytes in variant B were only represented by genera Cladosporium and Penicillum and in small measure.
Možnosti ovlivnění mikrobiologické čistoty potravin pomocí léčivých rostlin

Object of this thesis was to observe the microbiological purity of the juice and possible influence on purity by using extracts from selected species of medicinal, aromatic and culinary plants (MAPs). Presence of groups of Clostridium, Bacillus, E. coli, Enterococcus, Pseudomonas, molds and yeasts, psychrotrophic microorganisms and the total bacterial count was evaluated. Extracts of these kinds of plants were added to opened juice containers: fennel (Foeniculum vulgare var. dulce, L.), chamomile (Matricaria recutica, L.), hop (Humulus lupulus L.) and caraway (Carum carvi L.). Very good results after adding the extracts were achieved in growth restriction of groups of Clostridium, Bacillus, E. coli, Enterococcus and Pseudomonas. The extracts did not affect the growth of psychrotrophic microorganisms, molds and yeasts.
